Laboratory : Institut CARMeN-Caen (Team: Pr. Thierry Lequeux)
Title :Synthesis of new series of c-di-AMP analogs for antimicrobial activity.
Abstract : Given the rise in antibiotic resistance, there is an urgent need to develop alternative therapeutic options. The mechanisms of action of traditional antibiotics place stress on bacteria, thereby promoting the development of resistant strains. The development of new active ingredients with different mechanisms of action is therefore essential to combat these pathogens while minimizing the emergence of resistant strains. In this context, the important role of c-di-AMP, a secondary metabolite involved in the virulence and survival of pathogenic bacteria, makes it a promising target for the development of new anti-infective agents. Consequently, the development of inhibitors of diadenylate cyclases (DACs) and/or phosphodiesterases (PDEs)—enzymes responsible for the synthesis and degradation of c-di-AMP—appears highly promising for modulating the intracellular concentration of this metabolite and thereby affecting bacterial survival and/or virulence. In this thesis project, the design and synthesis of c-di-AMP analogs possessing high metabolic stability as well as good cellular permeability will be addressed to develop specific PDE and DAC inhibitors. To this end, new series of fluorinated analogs will be considered, for which it will be necessary to develop new synthetic methodologies.
Application : The candidate must have a strong background in synthetic organic chemistry. Preference will be given to candidates with experience in fluorine chemistry and/or multi-step synthesis and/or the chemistry of sugars and/or nucleosides. The candidate must demonstrate strong motivation, enjoy working in a team, and must have excellent skills in French (written and spoken) .
