Offer Description
The project focuses on the discovery and optimization of new compounds that interfere with the lysine and meso-diaminopimelate (DAP) biosynthesis pathway, with the aim of establishing a novel therapeutic strategy against bacterial pathogens. Particular attention will be given to the enzyme DapE (EC 3.5.1.18), a crucial and essential metalloprotease in this biosynthetic pathway (1)–(3).
In the search for potential inhibitors, one strategy—so far underexplored in DapE inhibitor design—is to develop compounds that mimic the natural substrate of DapE, LL-SDAP, or compounds exhibiting structural similarity to the tetrahedral intermediate formed during peptide bond cleavage by the protease.
- (1) Giese-type alkylation via photocatalytic decarboxylation;
- (2) Nickel-catalyzed alkene hydroalkylation;
- (3) alkene difunctionalization (carboimination) via energy transfer (EnT).
